A diversion is in place, and drivers are being warned to expect delays in the area.
Queues are currently backing up towards the M66 for drivers heading north from Bury and Manchester.
A spokesperson for National Highways said: "The A56 is closed in both directions between the A682 and the A680.
"Lancashire Police lead the response to the ongoing incident and have closed the carriageway to allow them to work at scene. National Highways have resources in attendance to assist with traffic management.
"Traffic is being diverted via the A681 and the A680. Delays are likely on approach to the closure and diversion routes.
"If this closure impacts on your planned route, please allow extra journey time. Plan ahead, you may wish to re-route or even delay your journey."
The closure follows an incident which was first reported at around 9.15pm, according to the Inrix traffic monitoring system.
An update from Inrix at around 10.20pm said: "A56 in both directions closed due to police incident between M66 J1 (Ramsbottom, Bury) and A680 Manchester Road (Bent Gate Roundabout, Townsend Fold)."
It reopened at about 11pm, police said.
