Centre-back Luiz gave a soft penalty away in the first half, being shown red after clipping Willian Jose as he ran through on goal.
It allowed Wolves to equalise after Nicolas Pepe's opening goal and Joao Moutinho then fired the hosts in front shortly after half-time with a fine long-range strike.
Things went from bad to worse for the Gunners when they were reduced to nine men in the 72nd minute when goalkeeper Leno inexplicably handled the ball when trying to clear outside the area under pressure from Adama Traore.
It effectively ended Arsenal's hopes of mounting a comeback as they suffered an unexpected defeat to an underperforming Wolves side.
Arsenal will now also be forced to deal without both Luiz and Leno for their next game against Aston Villa at the weekend.
The Gunners' subsequent game is against Leeds United, but will the Whites be handed a boost by the unavailability of two key opposition players for the match?
It seems unlikely.
Both players were sent off for denying goalscoring opportunities, which counts as a one-match suspension, which means the duo should be back to face the Whites.
It was the first time either player has been sent off this season, meaning neither will likely receive an additional suspension on top of their respective one-match bans.
