Valencia suffered a blow to their hopes on matchday 3 as Young Boys held them to a 1-1 draw in Switzerland, but the Spanish side can leapfrog United in the group - temporarily, at least - with a win at the Mestalla.
With Juventus likely to qualify top of the group and hosting United in Wednesday's late kick-off, Valencia have the perfect chance to lay down the gauntlet to Jose Mourinho's Red Devils.
