The fire started at a building in the city of Talca, which is about 158 miles south of the capital Santiago, at about 10pm on Tuesday.
Firefighters are battling the blaze and are said to be still searching for more people who may be trapped inside the building.
Horrifying footage has been shared on social media appearing to show flames billowing out from the building, with the roof charred.
Talca Fire Department officials said that tragically four people had died in the blaze, including two children.

They have not been identified but reports claim they are of Venezuelan nationality.
Local media reports that fire chiefs said the intensity of the blaze made it impossible to save those inside the building.
Eight fire crews are said to be at the scene bringing the fire under control.
While the cause of the blaze is still under investigation reports say that a preliminary view is that of an explosion from a gas cylinder.

While it has not been confirmed some reports say that the building was a shelter for migrants.
Talca's chief prosecutor Hector de la Fuene arrived at the scene to assist with the incident. Talca Fire Department are also starting an investigation into the cause of the blaze.
