The Belgium defender is a surprise absentee from the squad, and is reported to have picked up an injury, as Mauricio Pochettino makes four changes.
Eric Dier, Juan Foyth, Davinson Sanchez and Ben Davies come into the side.
West Ham made two changes with Ryan Fredericks and Issa Diop replacing Pablo Zabaleta and Angelo Ogbonna.
