The Belgium international, who has not featured for Spurs since their 2-0 victory over Huddersfield Town on September 29, has been sidelined with a hamstring injury but his return to the bench at the weekend provided a timely boost for Spurs.
Mauricio Pochettino's men head into a must-win Champions League clash with Inter Milan before Sunday's North London derby and Vertonghen is in line to end his long wait for an appearance.
With Davinson Sanchez (hamstring) unavailable, additional options at centre back will be well received.
Elsewhere, Mousa Dembele (ankle) and Kieran Trippier (groin) are also sidelined.
