The Seminoles led the entire game, allowing the Cardinals to pull within one point three times during the second half, but made timely free throws down the stretch to secure the victory.
Sophomore forward Terance Mann added 15 points and fifth-year center Michael Ojo added 10 points for the Seminoles, who thanked fans in the student section after the game.
The Seminoles finished their torrid six-game stretch against top 25 opponents with a 5-1 record, including four wins at home.
The win marked the first time ever FSU beat Louisville in Atlantic Coast Conference play.
FSU begins a three-game road trip at Georgia Tech on Wednesday.
