Greater Manchester Police said it was called to reports a person had fallen from the Britannia Hotel, in the city centre, at 5.30am on Wednesday.
A 20-year-old man was pronounced dead at the scene.
Officers gained entry to a room in the hotel, where they found a man’s body.
A witness said there appeared to be glass the road where the first man was found and a ripped curtain billowing from an upper-floor room at the seven-storey building.
Police have erected a forensic tent in Silver Street, at the rear of the hotel.
Detective Chief Inspector Amber Waywell, of the force’s major incident team, said: “We are currently working to establish exactly what has happened to lead to the death of both these men.
“We have a team of officers at the scene, and Silver Street in the city centre will remain closed as we carry out our investigations.
“If you have any information related to this incident, no matter how small or insignificant it might seem, I would urge you to get in touch with police.”
The Britannia is a Grade II*-listed Victorian building with 363 rooms. The former textile warehouse was converted into a hotel in 1982.
