The five-time winners, who last lifted the trophy in 2015, qualified for the first knockout phase of this year's competition by topping Group B ahead of Tottenham, Inter Milan and PSV Eindhoven.
Ernesto Valverde's La Liga leaders face a Lyon side that finished as runners-up to Manchester City - whom they beat in September - in Group F after holding Shakhtar Donetsk to a crucial 1-1 draw in Kiev in December.
OL, coached by former midfield stalwart Bruno Genesio, are currently third in Ligue 1, four points shy of Lille and 16 adrift of Paris Saint-Germain despite beating the latter earlier this month.
The hosts will be without talismanic captain Nabil Fekir through suspension and could face a reunion with former Lyon centre-back Samuel Umtiti, who is fit again after missing three months with a knee injury.
