Conor Murray and Tadhg Beirne are among 14 members of the Lions touring party isolating at the team's hotel after two positive Covid-19 tests today.
Last-minute Covid testing took place ahead of Wednesday night's game against the Sharks after it was revealed earlier that day that one member of management tested positive for the virus.
And the further tests showed another positive result for an unnamed player, with eight other players and four staff members deemed close contacts.
The situation forced Lions manager Warren Gatland to make wholesale changes to his squad for the match against the Sharks.
Among other changes, Tadhg Beirne, Conor Murray, Chris Harris and Stuart Hogg have been pulled off the bench, with Russell, Maro Itoje, Hamish Watson and Taulupe Faletau their replacements.
In total 14 members of their touring party are now isolating at the team's hotel in Johannesburg.
