Police in Tasmania have seized the biggest haul of cocaine in the state's history from a logistics truck that had allegedly undertaken a return trip to Victoria.

Some 1.4 kilograms of cocaine, 4.6 kilograms of cannabis and 392 grams of methylamphetamine were discovered inside the cabin of the truck on Tuesday.

It was pulled over by police on a highway in Hobart following a "long-running" investigation.

"The 1.4 kilogram seizure of cocaine is indeed Tasmania's largest seizure of cocaine ever," acting commander Natasha Freeman told reporters.

"That is quite significant. We're very pleased to get that off the streets."

Police allege the truck sailed to Victoria on the Spirit of Tasmania on February 27 and returned to the island on March 1.

A 60-year-old man from Brighton and a 58-year-old man from Elderslie have been charged with trafficking in a controlled substance.

They are expected to appear in Hobart Magistrates Court on Wednesday.

Police say the combined value of the drugs is between $700,000 and $1.2 million.
