The Dubliner was superior as she out-boxed Englands Caroline Dubois in Paris, earning the win on four of the five judges' scorecards.
An emotional Harrington celebrated with her coaches afterwards, with the win assuring she'll earn a high seeding in this summer's showcase.
Harrington was comfortable as she landed the majority of scoring shots against the 20-year-old on the way to her biggest result since becoming world champion in 2018.
As a wrist injury kept her out of the ring for much of 2019, and the Covid-19 outbreak wreaking havoc on her 2020 plans, Harrington is now Ireland's leading prospect to take home a boxing medal from Tokyo.
Elsewhere, Harrington's quarter-final opponent and professional IBF world pro champion Maiva Hamadouche sealed her ticket to the games after a win in a box-off of the 60 kg division.
In Tuesday's final session, Belfast's Michaela Walsh lost out on gold against Imra Testa from Italy in the featherweight division.
