Barca have already qualified for the knockout stages as Group F winners and coach Ernesto Valverde was expected to rotate his players for the match at San Siro.
The Argentine, who scored a hat-trick in Barca's 5-2 win over Mallorca on Saturday night, has been left out of the Catalans' 20-man squad, which includes a number of younger players.
Luis Suarez is likely to lead the line as he seeks his first away goal in the Champions League since 2015, but Messi’s absence means there could be a chance for Carles Perez on the right.
There could also be an opportunity for 17-year-old Ansu Fati, who is available again after a knock and has been named in the 20-man list for the game in Italy.
Neto is in line to make his debut in goal after signing from Valencia in the summer, while Samuel Umtiti should also come in for a start at the back as Gerard Pique sits out.
Sergi Roberto has also been stayed behind in Barcelona and with Nelson Semedo out, that means Moussa Wague will start. Another young right-back, Dani Morer, is also in the squad.
In midfield, Sergio Busquets is just one yellow card away from a ban and a booking would see him miss the first leg of Barca’s last-16 tie, so he is likely to be rested for this one.
Arturo Vidal, who is interesting Inter ahead of the January transfer window, is set to start in midfield and Tuesday night could also see an opportunity for Carles Alena after the Catalan came on at the weekend against Mallorca for his first minutes since August.
Talented 20-year-old midfielder Riqui Puig could play a part after being named in the squad, while young central defender Ronald Araujo is included along with Jean-Clair Todibo.
Ousmane Dembele misses out through injury, while Jordi Alba is not included despite returning to training this week.
Inter are second in Group F, level on points with Borussia Dortmund, and need to match the German side’s result to advance to the last 16.
Antonio Conte’s side have a better head-to-head record in the matches played against BVB and will go through if the two teams finish level on points. Dortmund are at home to Slavia Prague, with nothing to play for, in their match.
