The 150 Australians arriving in Canberra from Singapore on Thursday will be tested four times while in hotel quarantine.
Health officials will test passengers for the virus on day one, 10, 11, and 12 during their 14-day stay. Thursday's flight is the first since June, and there is expected to be one more repatriation flight to Canberra before Christmas.
The ACT Health Minister Rachel Stephen-Smith said the location of the hotel would not be made public to avoid a "media circus", however, she admitted the word would get out eventually.
"We want people to arrive with dignity and privacy," Ms Stephen-Smith said on ABC Canberra.
ACT Policing and the Australian Defence Force will give their support during the quarantine process.
Following Australian Health Protection Principal Committee advice, hotel staff and government employees in contact with returned travellers will be regularly tested for COVID-19 symptoms.
The ACT Health Minister said hotel staff were well prepared for the job ahead, and that ACT Health had identified staff with high-risk second jobs.
She said it was not necessary to pay hotel staff more to compensate for missing shifts at other workplaces.
She said there were already 118 people in hotel and home quarantine in the ACT.
There is still one active case of COVID-19 in ACT after a diplomat arrived back in Canberra last week.
There were no new cases reported on Tuesday.
