John Stones also bagged a brace from a pair of set piece headers in a goal-filled first half with Jesse Lingard also firing a sublime third either side of two Kane penalties.
The captain's third came via a deflection before Panama grabbed a late consolation to record some history of their own - their first-ever World Cup goal.
England now face Belgium in Kaliningrad on Thursday with top spot in Group G on the line.
